What is T4 Gene 32 Protein (T4 gp32)?
T4 gp32 is a specific single strand DNA binding protein that is involved in T4 bacteriophage replication and repair. T4 gp32 binds and stabilizes single stranded DNA and is utilized as an additive in molecular biology assays to increase DNA yield.
What are the applications for T4 gp32?
- PCR
- Electron microscopy
- Restriction enzyme digests
- RT-PCR
- Whole genome amplification (WGA)
- Helicase-dependent amplification
What is the storage buffer for T4 gp32?
T4 gp32 storage buffer is 20 mM Tris-HCl, pH 8.0, 1 mM EDTA, 100 mM NaCl, 50% Glycerol.
How do we assess concentration of T4 gp32?
T4 gp32 is supplied at a concentration of 10 mg/mL. Concentration is assessed by nanodrop (DeNovix), blanked using the storage buffer and an extinction coefficient of E1% 11.76 g/100 mL.
How do we assess the functionality of T4 gp32?
The ssDNA binding affinity of T4 gp32 (reported as Kd) is determined by incubating a dilution series of T4 gp32 with a constant amount of fluorescently labeled ssDNA. The resulting signal change in fluorescence polarization is used to determine the binding affinity (Kd).
What is the extinction coefficient for T4 gp32?
The extinction coefficient of E1% is 11.76 g/100mL.
